You know, I thought this was the case as well because most paylines go through the center spot on reel 3.  However,  compare the number of lines that cross all three spots on the center reel to any other reel.  In a 9 line game, there are nine lines that pass through each real so I am not sure if there is any advantage to holding the center card.  Once you hold a card, all three spots on the reel are covered and there are 9 lines that pass through that reel regardless of where that reel is located.
 
My wife believes that holding a pair on reel 1 and 5, the bookends, is the worst option.  I don't agree based on my comments above, but based on results at the casino, it seems this might be the case.  I certainly don't have enough data to prove it.
 
I have been on both sides of this so I hope an expert resolves this once and for all.

No, if you hold a card then every single line hits it. The number of lines would only vary on draw cards.

Yes, the center spot on the middle reel is more important in spin poker (only assuming you are playing 4 lines or more) because more potential winning combinations run through it. You have to conceptualize that spin poker is really part poker machine and part slot machine - even though the payouts are based on a poker machine's paytable and the machine is not programmed to payout at a certain rate (like a slot) but on the true odds of a 52 card deck.

However, unlike in a regular triple-five-ten play machine, not only what cards are revealed - but where they show up - also plays a factor in payouts.

In a three-five-ten play game - if 4 aces show up - it doesn't matter what position they come in - you are a guaranteed winner. In spin poker, 4 aces can show up or "be on the board" and if you are playing a nine line game you could possibly win 3 times, 2 times, one time, or zero times - depending on how many you were originally dealt, what card position they were dealt in, and exactly what final position those 4 aces are in - based on the standard 9-line configurations that multi-line slots use.

That is what can be really frustrating about spin poker. Many times you will see all 4 aces out there - yet, you often will not cash one 4oak because of the position they are configured in.
